How is this a vote against EU and euro? Does this indicate Italy's exit from the EU or eurozone?

Not at all. Referendums on international treaties are not allowed in Italy. The vote could indicate a rise of the populist leaders (Beppe Grillo of M5S, Matteo Salvini of North League), who usually make remarks and slogans against EU and euro.
